Abstract

ABSTRACT About 34 isolates of filamentous fungi as impacted local by petro derivatives in Suape, Pernambuco, Bahia and Pedra Branca, being investigated for enzyme production lignolitic system using diesel oil as substrate. A selection of fungi producing polyphenol oxidase was performed using the malt agar medium supplemented with gallic acid (0.5%). The fungi selected were transferred to Erlenmeyer’s flasks containing solution Manachini and diesel oil (1%), incubated at 30°C for 72 hours, and was analyzed: biomass production, pH and enzymes laccase, lignin peroxidase and manganese peroxidase by oxidation of ABTS, veratryl alcohol and phenol red, respectively. The results showed that only 12 fungi were able to produce polyphenol oxidases. The selected fungi grown on diesel formed higher biomass for the sample F9 (10.21 g / L + 0.2), with a pH ranging from 6.0 to 6.8 were observed. We detected the production of three enzymes in all samples tested, however, the highest production of laccase (290U/L-1), lignin peroxidase (144 U/L-1) and manganese peroxidase (60 U/L-1) respectively, occurred with the samples F33 and F25. The results showed that the biotechnological potential of fungi is related to impacted sites by petro derivatives, which favoring the isolation of microorganisms with ability to produce enzymes of lignolitic system.
