Lipase partition from Pseudomonas cepacia by aqueous phase-system PEG4000/phosphate

This work aimed to purify the lipases from by PEG 4000/phosphate ATPS. A system based on the equilibrium of the poly(ethylene glycol) (PEG) and phosphate salts was used. An experimental planning had done to evaluation of effects of pH (6, 7 and 8) and tie lines (PEG and salt equilibrium concentrations) on the partition coefficient. Total volume systems were of about 7 mL. Lipases were obtained from meddle contains soy oil by fermentation using Pseudomona cepacia yeast. The combination of the best date optimized partition and enzymatic activity was shown to pH 6.0 of ATPS PEG 4000/ phosphate as the best system for being utilized in the recovery of the lipolitics enzymes.Downloads
